Mazzon E, Cuzzocrea S. Role of TNFin ileum tight junction alteration in mouse model of restraint stress. Am J Physiol Gastrointest Liver Physiol 294: G1268–G1280, 2008. First published February 28, 2008; doi:10.1152/ajpgi.00014.2008.—Restraint stress induces permeability changes in the small intestine, but little is known about the role of tumor necrosis factor (TNF)in the defects of the TJ function. In the present study, we used tumor necrosis factor-R1 knockout mice (TNF-R1KO) to understand the roles of TNFon ileum altered permeability function in models of immobilization stress. The genetic TNFinhibition significantly reduced the degree of 1) TNFproduction in ileum tissues; 2) the alteration of zonula occludens-1 (ZO-1), claudin-2, claudin-4, claudin-5, and -catenin (immunohistochemistry); and 3) apoptosis (TUNEL staining, Bax, Bcl-2 expression). Taken together, our results demonstrate that inhibition of TNFreduces the tight junction permeability in the ileum tissues associated with immobilization stress, suggesting a possible role of TNFon ileum barrier dysfunction.

c-Jun NH2-terminal kinase-2 mediates osmotic stress-induced tight junction disruption in the intestinal epithelium.
Gastrointestinal epithelium faces osmotic stress, both at physiological and pathophysiological conditions. JNK activation is an immediate cellular response to osmotic stress. We investigated the effect of osmotic stress on intestinal epithelial barrier function and delineated the role of JNK2 in osmotic stress-induced tight junction (TJ) regulation in Caco-2 cell monolayers and ileum of Jnk(-/-...

In the present study, we used tumor necrosis factor-R1 knock out mice (TNF-alpha R1KO) to understand the roles of TNF-alpha on epithelial function in models of carrageenan-induced acute lung inflammation. In order to elucidate whether the observed anti-inflammatory status is related to the inhibition of TNF-alpha, we also investigated the effect of etanercept, a TNF-alpha soluble receptor const...
متن کاملSpinacia oleracea L. extract attenuates hippocampal expression of TNF-α and IL-1β in rats exposed to chronic restraint stress
Background: Restraint stress causes inflammation in nervous system that leads to emersion of neurodegenerative diseases. Spinach (Spinacia oleracea L.) contains different agents with antioxidant, antiapoptosis, and hepatoprotective properties. This study examined the effect of spinach hydroalcoholic extract (SHE) on TNF-α and IL-1β expression in hippocampus of male Wistar rats exposed to chroni...
متن کاملEffects of trichostatin A on pre- and anti-inflammatory cytokines of the hippocampus in rats of the prenatally restraint stressed model
Background: Prenatal Restraint stress (PRS) leads to cognitive-behavioral disorders and neuro-pathological deficits in children. The present study evaluated the effect of trichostatin A (TSA) on hippocampal level of pro- and anti-inflammatory cytokines in rat model of prenatal restraint stress. Materials and methods: In this experimental study, 20 pregnant Wistar rats were randomly divided into...
متن کاملEffect of Chronic Restraint Stress on HPA Axis Activity and Expression of BDNF and Trkb in the Hippocampus of Pregnant Rats: Possible Contribution in Depression during Pregnancy and Postpartum Period
Introduction: Brain-Derived Neurotrophic Factor (BDNF) and its receptor, TrkB, in the hippocampus are targets for adverse effects of stress paradigms in addition, BDNF and its receptor play key role in the pathology of brain diseases like depression. In the present study, we evaluated the possible role of hippocampal BDNF in depression during pregnancy, Methods: To achieve the purpose, repeat...
